There is a process to go through when reassembling. It's hard to know what component isn't working.
Blow in the fuel inlet hole before you put the bowl back on, and lift the float. did the valve stop the oncoming air? If so, assemble the bowl.
With the fuel screw out, blow compressed air into that passage. You will feel it coming out into the carb bore. Reassemble with spring, washer, and oring. A dad of grease to keep that assembly together long enough to get it started is perfectly acceptable. Tighten down until it seats lightly, back out 1 3/4 turns. There is often a chunk of ethanol sludge at the top of the fuel screw passage. I like to gently scratch the top of that area with a metal dental pick, and blow it out again.
Blow air into the pilot passage. Do you hear and feel air coming out of other areas of the carb? This is the number one cause of bikes not starting.
Does the accelerator pump shoot a solid blast into the intake passage? There is an adjustment for that, but right now, we're just trying to get it to start.
Will it start with the air filter removed and some starting fluid sprayed into the back of the carb with the seat removed? I don't like starting fluid as a crutch, but when we're trying to diagnose a no start condition, it is useful.
I have had bikes that I towed behind a car, holding onto a rope, with the choke out, pull it up into second gear and popped the clutch. Not textbook, but we want to get it to start, and then figure out why it is hesitant.
