What is your current elevation and jetting ??
Have you checked the valves to see if they may be closing up ???
Generally for cold starting (depends on the bike and jetting , as there are always variables) is you twist the throttle 5-6 times to load the fuel in the venturi ...slowly kick the bike over till you feel the compression at its highest , then release the kick stat and give it a full hard kick (usually with the choke on) , the bike should or almost start , if it does not start repeat the process and it should start right up .... I have found this to be the best cold procedure is to prime the carb so to speak with at least 5 but no more than 8 full twists , then get it on compression , and a manly kick ... this has worked for me for years as being the most reliable way to start these bikes when cold/first starting for the day ... usually afterwards just start them ...
Jetting Chart below , is where you should be
Jetting Guide for Main Jet:(pilot 45) CRF450R/X ... Fuel Screw should be no more than 3 turns out and no less than 1 in , otherwise go to next larger/smaller pilot jet ... Needle position is usually best around 3rd to 4th from top depending , for fine tuning of jetting , bogging , etc.

2000 ft.......162
4000 ft.......160
6000 ft.......158
8000 ft.......155
Temperature/Elevation:
One main jet size (up/down) for every 2,000 feet or 25 degrees in temp.
